Blood Group Test Procedure & Importance Explained
A blood group test is essential during life-saving blood transfusions. Through a simple blood group test procedure, your blood type will be classified as one of the eight blood types. The procedure is also known as ABO typing or Rh typing. Today, most pathology and medical laboratories offer the test at affordable costs. They also deliver the results within 12 hours. With a blood group test kit, you can also do it at home.
Knowledge of your blood group is indispensable these days. Whether you want to donate blood or receive blood, it's crucial. This article will cover the lab procedure for blood group tests. It will also discuss general test prices across the country.
Blood Group Test Procedure
The procedure of the blood group test is very straightforward. It doesn’t involve many technical details. So, you can also do it at home without endangering yourself.
Preparation for Blood Typing Test
Medical standards prescribe preparatory steps for patients seeking blood group tests.
- Always wear loose clothing that makes access to your arm veins easy.
- Before the test, tell your healthcare provider about your health history. Also tell them about past blood transfusions, organ transplants, and pregnancies (for women). These factors might affect your blood test results. They can help your provider care for you better.
- There is no fasting required for blood group tests.
Blood Sample Collection
The procedure for the blood group test is very simple and painless. You will also get the blood group test report on the same day or within 12 hours. The procedure of blood group test involves the following steps:
- The lab technician, a phlebotomist, will clean the area around your arm. They will tie a tourniquet on your upper arm to make the veins appear.
- The phlebotomist will now insert a sterilised needle into a vein to draw a small amount of blood.
- After collecting the blood, the phlebotomist will use a gauze pad to apply pressure at the area of the pinch. Then they will apply a cotton or bandage.
- Now, we will send the collected blood sample in the test tube to the laboratory to determine the blood group.
The whole procedure of blood sample collection should take, at most, a few minutes. As mentioned, you can expect the blood group test report on the same day.
Blood Grouping Principle and Procedure
Finding the blood group of a patient from the blood sample involves three simple steps: Forward Typing, Reverse Typing, and Rh-typing. The basic principle of ABO typing is the fact that the results of forward typing must match the results obtained in the reverse typing test with respect to antigens on RBCs and antibodies in blood plasma.
A blood group test kit, which contains reagents to detect the presence of antigens on the surface of RBCs, is used for this purpose.
Forward Typing
Mix the blood sample with antibodies against type A and type B blood. Observe the mixed sample for the formation of clumps, known as agglutination. Clumps show that the blood sample is reacting with the antibodies.
If a blood sample forms clumps when mixed with type A antibodies, its group is type A. If it reacts with type B antibodies, it's type B.
If your blood sample reacts with both antibodies, its typing is AB. If no clumping occurs with both types of antibodies, then your blood group is O.
Reverse Typing
The plasma of the blood sample is mixed with known blood types (A or B) in this procedure of blood test. Blood plasma of type A contains antibodies against type B blood and vice versa. If agglutination happens when mixed with type B, then your blood group is A; If agglutination occurs when mixed with type A, then your blood typing is B.
If a reaction occurs when mixed with both blood types, then that blood group is determined as O.
Rh-D Typing
The presence of the Rh (Rhesus) factor is determined in both forward and reverse typing procedures. During forward typing, blood plasma mixes with anti-Rh serum or type D antibodies. In reverse typing, the plasma of the blood sample mixes with Rh-positive red blood cells.
Combining forward and reverse typing results determines the patient's blood type. This reveals the blood group (A, B, AB, or O) and the presence of the Rh-factor.

Types of Blood Groups
What classifies different types of blood groups? It's the presence or absence of specific types of antigens on the membrane of red blood cells. There are two broadly classified blood groups: ABO and Rh.
In ABO, there are four subtypes A, B, AB, and O. Whereas, any blood group can be Rh positive or Rh Negative, which means there are a total of eight blood groups.
Common Blood Groups
| Type of Blood Group | Antigen on RBCs | Antibody in Plasma |
|---|---|---|
| A | A | B |
| B | B | A |
| AB | Both A and B | Nil |
| O | Nil | Both A and B |
*Each blood type can be Negative or Positive depending on the presence of Rh-D antigen on RBCs.
Why Blood Group Testing
ABO typing knowledge is crucial during blood transfusions. It prevents foreign blood types from entering one's body. Mixing blood types leads to severe immunological responses in the human body. These responses are often fatal. Recipients' blood cells release antibodies that fight foreign blood cells causing a toxic response. That's why medical professionals need to determine blood type before transfusions. This can avoid lethal consequences.

Conclusion
As mentioned, the procedure of blood group tests is simple and intuitive. All you need is a blood group test kit containing monoclonal antibodies (A, B, and D). Also, you'll need alcohol swabs, a cotton ball, a clean glass slide, toothpicks, and a lancet. Bring your blood sample, too. The best part? It is affordable. But, you can learn your ABO typing for free. Take part in blood donation programs and ask the staff.
If you're afraid of needles, there is another blood group test procedure. It uses saliva, mucus, and other fluids instead of a blood sample. Because 80% of people secret their blood group antigens into these fluids. All in all, ensure that you know your ABO typing beforehand.
